Penelitian bertujuan untuk mengkaji pengaruh kulit buah naga dan daun jambu biji dalam pakan sapi potong terhadap total protein cairan rumen, aktivitas enzim dan sintesis protein mikroba. Rancangan penelitian yang digunakan yaitu Rancangan Acak Lengkap pola one way classification. Pengukuran aktivitas enzim (amilase, selulase, protease) dan sintesis protein mikroba dilakukan pada inkubasi selama 4 jam, terdapat 3 perlakuan yang diuji yaitu terdiri dari P1 (Konsentrat 50% + jerami padi amoniasi 50% (Kontrol)); P2 (P1 + Kulit buah naga 5%); P3 (P2 + tepung daun jambu biji 2,5 %). Ketiga perlakuan diuji secara invitro. Setiap perlakuan diulang sebanyak 6 kali sehingga terdapat 18 unit percobaan. Selanjutnya analisa terhadap dinamika total protein cairan rumen dari masing-masing perlakuan dilakukan pada inkubasi 0, 2, 4 dan 6 jam dan ulangan 6 kali sehingga terdapat 72 unit percobaan. Hasil analisis variansi total protein cairan rumen menunjukkan bahwa perlakuan tidak berpengaruh nyata terhadap kontrol (P>0,05) pada inkubasi selama 0; 2; dan 6 jam akan tetapi hasil berpengaruh sangat nyata (P<0,01) pada inkubasi selama 4 jam, hasil kontras ortogonal menunjukkan bahwa terdapat pengaruh sangat nyata (P<0,01) antara substrat kontrol dengan yang ditambah kulit buah naga dan daun jambu biji (P1 Vs P2 - P3) terhadap penurunan total protein cairan rumen. Analisis variansi pada dinamika protein cairan rumen menunjukkan bahwa lama inkubasi berpengaruh nyata (P<0,05) terhadap kontrol (P1) dan substrat yang di tambah 5% kulit buah naga (P2). Hasil ortogonal polinomial terhadap dinamika protein cairan rumen menunjukkan bahwa lama inkubasi berpengaruh secara kubik terhadap total protein cairan rumen dengan persamaan P1 yaitu Y = 2.4978333 – 0.4305 dan koefisien determination (𝑅2) = 0,63, hasil persamaan P2 yaitu Y = 2.4201667 - 0.40406944 dan koefisien determination (𝑅2) = 0,67, demikian pula didapat total protein cairan rumen pada P1 dan P2 terendah terjadi pada inkubasi selama 1,4 jam sedangkan puncak total protein cairan rumen tertinggi terjadi pada inkubasi selama 4,9 jam. Hasil penelitian aktivitas enzim protease, amilase dan selulase menunjukkan bahwa perlakuan berpengaruh sangat nyata terhadap kontrol (P<0,01) dan hasil kontras ortogonal menunjukkan bahwa terdapat pengaruh antara kontrol dengan yang ditambah kulit buah naga dan daun jambu biji (P1 Vs P2-P3) namun berbeda pada aktivitas amilase menunjukkan bahwa terdapat pengaruh nyata (P<0,05) antara perlakuan yang ditambah kulit buah naga dan perlakuan yang ditambah kombinasi kulit buah naga dan daun jambu biji (P2 Vs P3). Pengamatan terakhir yaitu pada Sintesis Protein Mikroba (SPM) dengan hasil bahwa pakan perlakuan tidak berpengaruh nyata (P>0,05) terhadap pakan kontrol, namun nilai yang didapat dari ketiga perlakuan cenderung mengalami peningkatan total SPM. Berdasarkan penelitian yang telah dilakukan, dapat disimpulkan bahwa penambahan kulit buah naga 5% yang dikombinasi daun jambu biji 2,5% menurunkan total protein cairan rumen, akan tetapi meningkatkan aktivitas enzim (protease, amilase, selulase) dan sintesis protein mikroba rumen.

The study aimed to examine the effect of dragon fruit peel and guava leaves in beef cattle feed on total rumen fluid protein, enzyme activity and microbial protein synthesis. The research design used was a completely randomized one way classification pattern. Measurement of enzyme activities (amylase, cellulase, protease) and microbial protein synthesis were carried out at incubation for 4 hours, there were 3 treatments tested consisting of P1 (50% Concentrate + 50% ammoniated rice straw (Control)); P2 (P1 + 5% dragon fruit skin); P3 (P2 + 2,5% guava leaf flour). All three treatments were tested in vitro. Each treatment was repeated 6 times so that there were 18 experimental units. Furthermore, an analysis of the total dynamics of rumen fluid protein from each treatment was carried out at incubation of 0, 2, 4 and 6 hours and repeated 6 times so that there were 72 experimental units. The results of the analysis of the total variance of rumen fluid protein showed that the treatment had no significant effect on control (P> 0,05) on incubation for 0; 2; and 6 hours but the results had a very significant effect (P <0,01) on incubation for 4 hours, the results of orthogonal contrast showed that there was a very significant effect (P <0,01) between the control substrate with added dragon fruit peels and guava leaves seeds (P1 Vs P2 - P3) to decrease in total rumen fluid protein. Analysis of variance on rumen fluid protein dynamics showed that the incubation time had a significant effect (P <0,05) on control (P1) and the substrate added 5% dragon fruit peel (P2). The polynomial orthogonal results on the dynamics of rumen fluid protein showed that the incubation time had a cubic effect on the total rumen fluid protein with the P1 equation Y = 2,4978333 –0,4305 and the coefficient of determination (𝑅2) = 0,63, the result of the P2 equation Y = 2,4201667 – 0,40406944 and the coefficient of determination (𝑅2) = 0,67, as well as the lowest total rumen fluid protein at P1 and P2 occurred during incubation for 1,4 hours while the highest peak of total rumen fluid protein occurred at incubation for 4,9 hours. The results of the protease, amylase and cellulase enzyme activity showed that the treatment had a very significant effect on control (P <0,01) and the results of orthogonal contrast showed that there was an influence between the control with added dragon fruit peels and guava leaves (P1 Vs P2-P3 ) but different on amylase activity showed that there was a real influence (P <0,05) between the treatment added with dragon fruit peel and the treatment added with the combination of dragon fruit peel and guava leaves (P2 Vs P3). The last observation was on Microbial Protein Synthesis (SPM) with the result that the treatment feed had no significant effect (P> 0,05) on the control feed, but the value obtained from the three treatments tended to experience an increase in total SPM. Based on the research that has been done, it can be concluded that the addition of 5% dragon fruit peel combined with 2,5% guava leaf decreases total rumen fluid protein, but increases enzyme activities (protease, amylase, cellulase) and rumen microbial protein synthesis.
