Abstrak :
Ameloblastoma merupakan tumor odontogenik yang bersifat lokal invasif dengan pertumbuhan lambat. p16 bertindak sebagai penghambat interaksi antara cyclin D dengan CDK 4/6 serta menghalangi siklus pembelahan sel pada fase G1. Tujuan penelitian ini adalah menganalisis ekspresi p16 pada ameloblastoma tipe folikuler dan pleksiform. Jenis penelitian ini adalah deskriptif analitik dan teknik sampling menggunakan total sampling. Subjek penelitian sebanyak 8 sampel ameloblastoma tipe folikuler, 9 sampel ameloblastoma tipe pleksiform, dan 4 sampel kontrol normal gingiva. Pengecatan imunohistokimia sampel menggunakan antibodi p16 mutan. Hasil diinterpretasikan dengan skoring imunoreaktif. Hasil uji statistik didapatkan nilai rerata ekspresi p16 pada ameloblastoma tipe folikuler 2,13 ± 1,808; tipe pleksiform 4,44 ± 2,506, dan kontrol gingiva 0 ± 0. Hasil uji Kruskal wallis didapatkan nilai p=0,006 (p<0,05) menunjukkan terdapat perbedaan yang bermakna antara kelompok kontrol normal, ameloblastoma folikuler, dan ameloblastoma pleksiform. Hasil uji Mann-whitney menunjukkan ekspresi p16 pada kontrol gingiva berbeda nyata dengan ameloblastoma tipe folikuler (nilai p=0,075), kontrol gingiva berbeda nyata dengan ameloblastoma tipe pleksiform (nilai p=0,008), dan ameloblastoma tipe folikuler tidak berbeda nyata dengan ameloblastoma tipe pleksiform (nilai p=0,146). Simpulan dari penelitian ini adalah ekspresi p16 mutan tidak berbeda nyata pada kelompok ameloblastoma tipe folikuler dan tipe pleksiform sehingga tidak dapat digunakan sebagai petanda biologi molekul untuk mengetahui progresifitas ameloblastoma menggunakan pengujian imunohistokimia.

P16 act as a barrier to interaction between cyclin D with CDK 4/6 as well as blocking the cell division cycle in the G1 phase. The aim of this study was to describe the expression of p16 on follicular and plexiform type ameloblastoma. This research was analytical-description and use total sampling technique. The subject of this study used 8 sample follicular type ameloblastoma, 9 sample plexiform types ameloblastoma, and 4 samples normal gingiva. Immunohistochemical staining in sample was performed using p16 mutant antibody. The results were interpreted with immunoreactive scoring. The statistical test obtained mean value p16 expression in follicular type ameloblastoma was 2,13 ± 1,808; in plexiform type ameloblastoma was 4,44 ± 2,506; and in control group was 0 ± 0. Kruskal wallis test shown that there was a significant difference between the follicular, plexiform, and control group (p=0,006; p<0,05). Mann-whitney test shown that there were significant differences between the p16 expression in control group and follicular group (p=0,075) and between the control group and plexiform group (p=0,008). There wasn’t significant difference between follicular and plexiform type ameloblastoma (p=0,146). The conclusion of this study was p16 mutan expression was not significant difference between follicular type ameloblastoma to plexiform type ameloblastoma so they could not used as molecular biology marker to know progresiveness of ameloblastoma using immunohistochemistry testing.
